Intended Use

This kit can be used to determine the percentage and number of yeast and mold, bacteria, Brettanomyces, and total aerobic counts in a beer or wine sample.

Procedure

Mix and collect sample using provided sterile bulb pipette and transfer to sterile collection tube. Label tube “tube 1” (a sharpie pen works best for this)

Serial dilution:
a. Transfer 1 ml from tube 1 to a tube filled with 9 ml dilution buffer (provided) by dropping in solution from tube 1 until 10 ml is reached.
b. Cap and invert tube 6 times
c. Transfer 1 ml to next tube (same as step a) and label tube.
Continue dilution series for all 6 tubes.

Apply 40 µL using calibrated micropipette or, for semi-quantitative result, 1 drop from supplied bulb pipette directly to agar. To each of the four types of plates [WL, total bacteria, total yeast (and mold), and Brettanomyces (and mold)].

Add 1 tube’s worth of supplied glass beads, cover plate.

With plate flat on the table, slide/shake plate back and forth about 6-8 times.

Remove top and invert plate over waste container to dispose of beads. Replace top.

Incubate plates at 25ºC–30ºC in supplied self-sealing incubation bags for up to 2 weeks.
Interpretation

Total aerobic counts:

If single colonies develop, count and record the number of colonies. Multiply the number of colonies by 25 to get the number of colony forming units (CFU) per ml. If a haze develops over the entire plate and/or the plate turns yellow, your sample is TNTC (To Numerous To Count) and contains over 10,000 CFU/ml. If you get a TNTC result: Repeat the assay with serial 1:10 dilutions in sterile tubes using sterile buffer to determine the exact cell concentration.

Precautions
Use sterile technique whenever plates are open.

Minimize the time that the top is off the dish. Work in a draft-free environment.

Avoid directly touching the agar medium.

Seal plates within supplied self-sealing bags to reduce plate drying.

Storage
Store refrigerated between 2-8ºC.
Shelf life: 3 months
